ABSTRACT

Objective@#To investigate the diagnostic value of combined detection of carcinoembryonic antigen (CEA), carbohydrate antigen 199 (CA199) and carbohydrate antigen 125 (CA125) in lung cancer.@*Methods@#From July 2017 to February 2019, 64 patients with lung cancer, 40 cases of benign pulmonary lesions and 40 healthy people were selected by random lottery in Fuyang Hospital Affiliated to Anhui Medical University.Serum CEA, CA199 and CA125 levels of each group were analyzed.@*Results@#The levels of serum CEA, CA199 and CA125 in patients with lung cancer were significantly higher than those in patients with benign lung disease, the differences were statistically significant (t=18.355, P<0.001; t=11.757, P<0.001; t=10.754, P<0.001). The serum CA125 of lung benign disease group was significantly higher than the healthy group, and the difference was statistically significant(t=6.942, P<0.001). The positive rates of serum CEA, CA199 and CA125 in patients with lung cancer were significantly higher than those in the benign lung disease group and healthy group, the differences were statistically significant(χ2=22.110, P<0.001; χ2=13.321, P=0.002; χ2=15.125, P=0.001). The serum levels of CEA, CA199, and CA125 in patients with middle and advanced stage were significantly higher than those in the early stage, the differences between the advanced group and the early group were statistically significant (t=5.587, P<0.001; t=5.068, P<0.001; t=8.962, P<0.001). Those between the mid-term group and the early group had statistically significant differences (t=4.44, P<0.001; t=4.716, P<0.001; t=12.700, P<0.001). The CEA level of patients with advanced lung cancer was significantly higher than that of the intermediate patients.There was statistically significant difference in the CEA level between the advanced group and the mid-term group (t=3.860, P=0.001). Serum CA125 was the most sensitive in the diagnosis of lung cancer (60.84%), but had the lowest specificity (65.17%). CA125 compared with CA199, CEA, the difference was statistically significant (sensitivity: χ2=10.723, P<0.001 specificity: χ2=4.203, P=0.023). The sensitivity of CEA+ CA199+ CA125 combined detection was significantly higher than single detection, the difference was statistically significant(χ2=26.047, P<0.001),@*Conclusion@#The combined detection of serum CEA, CA125 and CA199 has high sensitivity in the diagnosis of lung cancer, which can improve the detection rate of early lung cancer and is worthy of clinical application.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a perceived fatigue evaluating model during simulated load carriage that is based on objective variables through analyzing the characteristics and trends of shoulder force, shoulder pressure, waist pressure, back pressure, and perceived fatigue, and to provide an analytical technique for research on load carriage.</p><p><b>METHODS</b>A 50-min simulated walking (at a speed of 5 km/h and a slope of 0%) experiment including 14 healthy male adults was conducted under four levels of backpack payloads (25, 29, 34, 37 kg). Shoulder force and trunk pressure were sampled simultaneously and analyzed with time- and frequency- domain methods. Multivariable linear regression was used to build a perceived fatigue evaluating model during load carriage.</p><p><b>RESULTS</b>The perceived fatigue evaluating model based on shoulder force, trunk pressure distribution ratio, load, and body mass index (BMI) was established. Its adjusted determination coefficient (aR2) was 0.709 and the absolute percentage error (APE) at the end of the experiment was less than 20%. The goodness of fit of the model based on frequency-domain independent variables was much higher compared with the model based on time-domain independent variables. The addition of BMI that represents the individual differences to the model obviously improved the goodness of fit.</p><p><b>CONCLUSION</b>The perceived fatigue evaluating model established in this study does not rely on the physiological changes of individuals, and thus can be used to establish an evaluation system for human load carriage with dummy as a substitution for human in experiments and to provide a scientific basis for efficient human load carriage.</p>

ABSTRACT

Aim To study the function of VR1 in chronic pain, to construct VR1 siRNA expression vectors and to study their silencing effect in the DRG neurons of rats were detected.Methods The hairpin sequences of siRNAs targeting VR1 gene of rat were designed, and two pairs of oligonucleotide sequence were synthesized. The annealed oligonucleotide fragments were cloned into linearized pRNAT-U6.2/Lenti expression vector and identified by PCR and DNA sequencing.Then, they were co-transfected by lipofectamine into 293T cells.The silencing effects of the lentivector-mediated VR1 siRNAs on the expression of VR1 mRNA were determined by RT-PCR after intrathecal injection in rats.Results DNA sequencing showed that the oligonucleotide fragments were correctly cloned into linearized pRNAT-U6.2/Lenti expression vector and the expression of VR1 mRNA in L4-L6 DRG neurons was inhibited significantly by pRNAT-U6.2/Lenti-siVR1 after intrathecal injection in rats.Conclusion The lentivector-mediated siRNAs are successfully constructed and they inhibit the expression of VR1 mRNA in the DRG neurons of rats, which may provide a potential tool for the further study and treatment of chronic pain.
